B1400282 [Abstract] OBJECTIVE: To investigate the cytochrome P4502D6 * 10 (CYP2D6 * 10) gene polymorphism and assess its effect on serum sex hormones, semen parameters and natural pregnancy rate in patients with idiopathic oligospermic male infertility treated with tamoxifen combined with testosterone undecanoate. METHODS: This case-control study included 230 patients with idiopathic oligospermic male infertility and 147 normal controls. The case group was treated with tamoxifen citrate, 20 mg/d, and testosterone undecanoate, 40 mg/d, for a total of 6 months. The CYP2D6 * 10 gene was typed using restriction fragment length polymorphism polymerase chain reaction (PCR-RFLP) technique. Sex hormone levels, semen parameters and spousal natural pregnancy rate of the study subjects were measured at the beginning of the study, in March and in June, respectively. RESULTS: There was no difference in the frequency of CYP2D6 * 10 C / C, CYP2D6 * 10 C / T and CYP2D6 * 10 T / T genotype distribution between the control and case groups (P = 0.707, P = 0.985, P = 0.751). At 3 months, folliculopoietin (FSH), luteinizing hormone (LH), testosterone (T) levels and semen parameters started to increase at 3 months and were significantly higher at 6 months. Statistical analysis revealed that in the CYP2D6 * 10 C / C group were significantly higher than C / T and T / T (P < 0.05). In addition, the spontaneous pregnancy rate of the patients' spouses was significantly higher in the wild-type (CYP2D6 * 10C/C) group than in the mutant group (CYP2D6 * 10 C/T and CYP2D6 * 10 T/T) (P < 0.05). CONCLUSION: Patients with CYP2D6 * 10 mutant idiopathic oligospermic male infertility received moxifen combined with testosterone undecanoate with poorer efficacy than the wild-type group.
