Implications of adding Huangqi Chi Feng Tang for IgA nephropathy

  To investigate the effects of Gao Wei Huang Qi Chi Feng Tang on the proliferation of glomerular thylakoid cells (GMC) stimulated by lipopolysaccharide (LPS), the expression of laminin (LN), collagen IV (Col-IV) and fibronectin (FN) in the cell supernatant and the cell cycle, and to explore its significance in the treatment of IgA nephropathy and prevention of renal fibrosis.  After gavage, drug-containing serum was prepared and GMC were cultured in DMEM containing 10% drug-containing serum and LPS was used as the stimulating factor, and the cells were divided into blank group, LPS group, temisartan group, and high, medium and low dose groups of Gavage Astragalus Chifeng Tang. Observe the proliferation of GMC under the effect of various treatment factors. ②The secretion of extracellular matrix (ECM), i.e., LN, Col-IV and FN, was measured by enzyme-linked immunoassay adsorption assay (ELISA) in GMC cultured in vitro under the effect of various treatment factors. (iii) Flow cytometry was used to detect the cell cycle.  Compared with the normal group, each drug-containing serum had no toxic effect on GMC (P>0.05); LPS stimulated the proliferation of GMC cultured in vitro, and the cell proliferation in the 0.625-80ug/ml LPS group was statistically significant compared with the normal group (P<0.05< span="">); the proliferation of GMC stimulated by LPS was inhibited in the group with added flavored Huangqi Chi Feng Tang at 24, 48 and 72 hours The inhibition rate of each group was the highest at 24 hours; at a fixed time point, there was no statistical difference between the medium-fold group and the high-fold group, and there was a statistical difference between the medium-fold group and the low-fold group (P<0.05< span="">); the high and medium doses of Radix Astragali Chifeng Tang blocked the cell cycle, increased the percentage of G0/G1 phase cells and decreased the percentage of S phase cells In the effect on ECM, the levels of Col-IV and FN were significantly reduced in the high-dose group (P<0.01< span="">) compared with the model group at 72 hours, while the levels of LN were not significantly changed in the medium-dose group (P<0.01< span="">). The contents of Col-IV, FN and LN were significantly reduced in the middle dose group (P<0.01< span="">); only the contents of FN were reduced in the low dose group (P<0.01< span="">), while the contents of Col-IV and LN did not change significantly.  LPS can promote GMC proliferation; Gao Wei Huang Qi Chi Feng Tang can significantly inhibit the proliferation of GMC stimulated by LPS and block the thylakoid cells in G0/G1 phase, and can reduce the secretion of ECM, indicating that GMC is an important target cell for the therapeutic effect of Gao Wei Huang Qi Chi Feng Tang and the prevention of renal fibrosis; Gao Wei Huang Qi Chi Feng Tang inhibits the proliferation of GMC with a certain dose- and time-dependent relationship. This indicates that the duration and dosage of our clinical medication are reasonable and that long-term medication is safe and effective.
