Formulation and specifications: Capsules: 12.5mg, 25mg, 37.5mg, 50mg
Indications: Advanced kidney cancer.
Key points for rational drug use:
1. The recommended dose is 50 mg orally once daily for 4 weeks and 2 weeks off (4/2 dosing regimen).
2. The dose is adjusted gradually in 12.5 mg gradient units based on individual patient safety and tolerability. The maximum daily dose should not exceed 75 mg and the lowest dose should be 25 mg. Treatment may need to be interrupted depending on the safety and tolerability of the individual patient.
3. Avoid combining with potent CYP3A4/5 inhibitors or inducers.
4. Severe myelosuppression such as decreased white blood cells and platelets may be observed after 4 weeks of sunitinib administration and 2 weeks of discontinuation (4/2 dosing regimen), so close monitoring of blood work is required during dosing. If necessary, a 2/1 dosing regimen can be used, i.e., 2 weeks of sunitinib and 1 week of discontinuation.
