Captopril is an effective anti-hypertensive drug. Recent studies have shown its effectiveness in promoting stem cell transplantation therapy. However, some studies imply its opposite effects in some cases. Mesenchymal stem cells (MSC) are commonly used for tissue engineering and pharmacological studies due to their pluripotent differentiation capacity and ease of amplification. In our study, we added different doses of captopril to isolate and culture newly born rabbit MSC. cell viability and explored the morphological characteristics of the dose-response relationship observed between captopril and MSC. newly born laboratory purchased Japanese White rabbits from the Medical Laboratory Animal Center, Sichuan, China. Bone marrow MSCs were isolated under sterile conditions. After 3 passages, MSC were trypsinized and re-suspended in α-MEM medium. 5×104/100ul cells were injected into each hole of 96-well plate. 20 ug/ml to 2400 ug/ml was added to a certain amount of captopril in three wells. 48h after culture, cell viability was detected by MTT method and morphological characteristics were observed. MSC grew well with newborn rabbit bone marrow. Endpoints, suspended cell morphology in the high-dose captopril group, each group showed distinctly different colors after 4h of MTT addition. The floating surface was absorbed in uncentrifuged and centrifuged at 500 g for 5 min under conditions and dimethyl sulfoxide was added in respectively. Absorbance values at 570 nm were then compared. The results show that high doses of captopril promote MSC proliferation, but not cell attachment. 100 – 300 ug/mL doses can ensure normal. Function and proliferation of MSC. The effect of captopril on MSC is still under debate. In our study, the divergence of doses of captopril may be one of the answers. Although doses of 100 – 300 ug/mLseems to be better for culture selection conditions, too high is the reality. We have reason to believe that the higher the dose of captopril, the better the facilitation effect of captopril in MSC. due to the convenience, rabbit MSC has the same value to explore and solve similar problems.
