Application experience of frozen section automatic staining machine

  With the rapid development of modern pathology technology, the frozen section staining machine is gradually accepted and used by major hospitals and related laboratories for its excellent performance and good section processing effect, especially its stability, efficiency and convenience, and gradually replaces the current manual staining; the application of this staining machine for intraoperative frozen section processing efficiency is significantly improved, the staining effect is stable The staining quality has been improved significantly. In the process of using the machine, the staining procedure and stain selection and replacement were compared and the most suitable solution was selected, which further improved the staining efficiency and staining quality.  1.Apparatus and equipment: Thermo (Sinisain) frozen section automatic staining machine and accompanying equipment.  2. Reagents: latest modified hematoxylin, modified differentiation solution, blue return solution, eosin, 85% ethanol, 95% ethanol, anhydrous ethanol, environmental protection transparent agent.  3.Staining: 3.1 Mount a conventional frozen section on a slide, fix it immediately, wash it with water, and mark the number of the section on the frosted surface of the slide, put it into the slide holder of the staining machine and place it on the conveyor belt of the staining machine, and prepare to start staining.  3.2 In practice, if the lymph node frozen section is stained, the first cylinder of hematoxylin solution can shorten the staining time appropriately, and the staining clip can be placed in the middle of the first cylinder; if a section takes a long time to stain in hematoxylin solution, the section can be put into the first cylinder first, and the power can be turned on after 30 seconds or 1 minute.  4. Precautions: 4.1 Before starting the power supply, the water of each flushing cylinder must be full. Due to the high pressure of water, the water flow valve must be adjusted as needed to control the water flow (after staining, the adjustment water flow valve must be closed to the minimum position).  4.2 The amount of dyeing solution in each dyeing tank can be executed with reference to the standard. (See Table II) (Hematoxylin, Eosin liquid liquid cannot be loaded too much, such as too much, in the water wash or into the next cylinder will not be able to clean the excess residual stain on the slide, which will lead to the pollution of the next cylinder stain and the unclean slides) 4.3 Keep the flushing cylinder clean. (The water outlet should be slightly lower than the first cylinder, and the outlet pipe should be as short as possible) 4.4 The mounting position of the slides should be appropriate, at 1/3 of the slide, not higher than that, such as higher may have an impact on the staining of the slides and the cleanliness of the whole slide, which will affect the final diagnosis of the pathologist.  4.5 After each staining, clean the staining machine and cover the staining cylinder cover to avoid contamination and evaporation of the staining solution.  Discussion: Frozen section, as a rapid diagnostic method, has long been widely used in routine pathological diagnosis. The accuracy of frozen section diagnosis is closely related to the high quality of frozen section and staining, but the frozen section stained by conventional staining methods (due to different configuration of reagents and staining methods) is not clear and transparent enough for microscopic observation, which often causes some difficulties for pathologists in diagnosis. This staining machine can stain several slides at the same time, thus saving a lot of time and allowing a clear view of the whole staining process of each slide; the improved staining solution used in the automatic staining machine for frozen sections has produced satisfactory results, and the sections are both clear and transparent under the microscope, almost the same as paraffin sections.
