Indications: Patients with unresectable hepatocellular carcinoma who have not received prior systemic systemic therapy.
Key points for rational drug use: 1. The most common adverse reactions during dosing include skin reactions on hands and feet, diarrhea, decreased platelet count, hypertension, elevated aspartate aminotransferase, alopecia, rash, and proteinuria.
2. The recommended dose is 0.2g per dose, twice daily, taken orally on an empty stomach and swallowed with warm boiled water. It is recommended to take the medication at the same time of day. If the medication is missed, there is no need to make up the dose and the next dose should be taken according to the regular dosing schedule.
3. In vitro studies suggest that Donafinil is mainly metabolized by CYP3A4 and UGTIA9, in addition to CYP1B1, CYP2C8, CYP2C9, CYP2C19, CYP2D6 and CYP3A5, which are also partially involved in the metabolism of Donafinil. Caution should be exercised when co-administering inhibitors or inducers of related metabolic enzymes.
