The effects and contraindications of domperidone tablets

Domperidone is a benzimidazole compound with strong pro-gastric motility and antiemetic effects, which can strengthen the tone of lower esophageal sphincter, prevent gastroesophageal reflux, enhance gastric motility, promote gastric emptying, coordinate gastric and duodenal motility, inhibit nausea and vomiting, and effectively prevent bile reflux independent of gastric juice secretion. It is a peripheral dopamine receptor antagonist, which directly blocks the dopamine receptors in the stomach and intestines, and presents a pro-gastric motility effect, which enhances peristalsis and lower esophageal sphincter tone. Because of its poor penetration of the blood-brain barrier and almost no antagonistic effect on dopamine receptors in the brain, mental and central nervous side effects can be excluded. The treatment of nausea and vomiting caused by Parkinson’s syndrome with dopamine receptor agonists is a special indication for this product. Contraindications for domperidone include hypersensitivity to domperidone, pheochromocytoma, breast cancer, mechanical intestinal obstruction, gastrointestinal bleeding, and pregnant women. Special attention should be paid to patients over 60 years of age or those taking more than 30 mg daily, which may lead to the risk of severe ventricular arrhythmias and sudden cardiac death, and this product should be used with caution in elderly patients or those with a history of heart disease.
