Ametinib Almonertinib

Formulation and Specifications: Tablets: 55 mg
Indications: Treatment of adult patients with locally advanced or metastatic NSCLC who have experienced disease progression on or after prior treatment with EGFR-TKI and whose presence of a positive EGFR-T790M mutation has been confirmed by testing.
Key points for rational drug use:
1. The recommended dose of ametinib is 110 mg orally once daily, either on an empty stomach or with a meal, swallowed whole, not chewed or crushed. For patients who cannot swallow the drug whole and need to be fed through a nasogastric tube, the tablets can be dissolved directly in carbonate-free drinking water and taken after complete dispersion.
2. The EGFR-T790M mutation must be clearly detected by a test approved by the State Drug Administration before administration.
3. Common adverse reactions to ametinib are rash, elevated blood creatine phosphokinase and pruritus, with a relatively low incidence of diarrhea. No interstitial pneumonia has occurred to date at the recommended dose, but care is still required during dosing.
4. Avoid combination with strong inducers and strong inhibitors of CYP3A4. Narrow therapeutic window drugs with breast cancer resistance proteins and P-glycoprotein sensitive substrates should be used with caution. Avoid combination with drugs that elevate blood creatine phosphokinase (e.g. statins).
