Formulation and specifications: Capsules: 150mg, 200mg
Indications:
1. Newly diagnosed adult patients with Philadelphia chromosome-positive chronic myeloid leukemia in the chronic phase.
2. Adult patients with Philadelphia chromosome-positive Ph+ CML in the chronic or accelerated phase who are resistant or intolerant to prior therapy (including imatinib).
Key points for rational drug use:
1. Nilotinib should not be used in patients with hypokalemia, hypomagnesemia, or long QT syndrome. Hypokalemia and hypomagnesemia must be corrected prior to nilotinib administration and monitored regularly.
2. Common adverse events are neutropenia, thrombocytopenia, anemia, loss of appetite, skin rash, and muscle and joint pain.
3. Avoid combining drugs that prolong the QTc interval and potent inhibitors of CYP3A4.
4. Dose reduction is recommended for patients with hepatic impairment.
5. Electrocardiogram should be performed regularly to monitor QTc before, during, and after initiation of dosing, and also during any dose adjustment.
