Advances in anti-tumor angiogenesis research of Chinese medicine

Tumor is a malignant disease that seriously endangers human health. The occurrence of tumor is increasing year by year compared with before. According to statistics: malignant tumor ranks lth among the causes of death of urban residents in China, Folkman found through his research that tumor growth and metastasis are vascular-dependent, and the current research on the mechanism of tumor vascular growth and inhibition of angiogenesis in tumor treatment is the research direction of most scholars. An important process of tumor growth is tumor angiogenesis, tumor cells are sent to the body circulation by new blood vessels, and tumors grow to target organs, forming tumor metastasis. When the tumor tissue grows to more than 2, mm3, it needs neovascularization to provide sufficient oxygen and nutrients for the tumor to continue to grow, and when neovascularization is lacking the tumor can be in a tiny state of 1-2, mm3 for a long time and then degeneration occurs. Therefore, tumor growth depends on angiogenesis. When a large number of tumor-related blood vessels grow inside the tumor parenchyma, it can promote continuous tumor growth or even metastasis; on the contrary, tumor growth will be significantly restricted and tumor cells will suffer from apoptosis or necrosis,. Tumor angiogenesis can be divided into the following steps: firstly, the relationship between tumor angiogenesis and inhibitory factors is imbalanced, and the phenotype of angiogenesis is generated; secondly, the degradation of vascular endothelial basement membrane occurs; the proliferation of endothelial cells in the process of tumor tissue migration and migration; the endothelial cells form a vascular ring after ductalization and branching; and finally, new basement membrane is generated. The current anti-tumor angiogenesis has become a direction of tumor therapy, and inhibitors of tumor angiogenesis will become a new field of research by inhibiting different aspects of tumor angiogenesis. Many current research findings have found that some single active ingredients of TCM as well as some TCM compound preparations can effectively inhibit neovascularization, thus inhibiting tumor appreciation and metastasis. The following analysis and summary are divided into single herbal medicines and compound herbal preparations. I. Research on single herbal medicine 1. Ginsenoside Ginseng is one of the representative drugs to support the essence of the body and has the efficacy of tonifying the vital energy and restoring the pulse and fixing the detachment. After the analysis of modern molecular biology technology, it was found that the main anticancer substance of ginseng is the diol-like tetracyclic triterpenoid saponin (Rg3) with the molecular formula C24H72O13. Ginsenoside Rg3 has obvious inhibitory effect on the growth of blood vessels in various tumor tissues. Sulforaphane is one of the early anti-tumor neovascularization drugs, which exerts anti-neoangiogenic effects by inhibiting basic fibrillogenic growth factor (bFGF), and Rg3 has similar anti-neoangiogenic effects as sulforaphane. Zhang Qingyuan et al. combined low-dose cyclophosphamide and Chinese ginsenoside Rg3 in tumor-bearing mice. It was observed that the mice in the combined group had the longest survival period and the vascular growth of tumor tissues was inhibited; the levels of microvascular density (MVD) and vascular endothelial growth factor (VEGF) in tumor tissues were decreased in the Rg3 group, and the decrease was more obvious in the combined group, indicating that ginsenoside Rg3 has an anti-angiogenic effect. Tao Houquan et al. used a rat in situ implantation model of gastric cancer tissue block as an in vivo test and found that Rg3 had a significant tumor growth inhibitory effect in vivo. Wang Jinliang et al. found that ginsenoside Rg3 could inhibit the angiogenesis of acute leukemia bone marrow by culturing bone marrow stromal cells of normal human and acute leukemia patients, and the progression of acute autohemorrhagic disease was delayed. In recent years, it has been found that Radix et Rhizoma also has good anti-tumor activity, and both Radix et Rhizoma lactone alcohol and erythropoietin are substances extracted from it, both of which have inhibitory effects on tumor neovascularization. Wang Guoping et al. demonstrated that rehmannia lactone alcohol could inhibit VEGF, mRNA expression and VEGF protein synthesis in human gastric cancer SGC7901 cells after 24h. Jinbo Zhu et al. used the self-established vascular endothelial cell line F2 and its serum-free culture sub-strain F2C to observe the effect of erythropoietin on the process of angiogenesis. The results showed that rehmannia could inhibit F2 and F2C wandering and proliferation, suggesting that it may be one of the important mechanisms for its induction of cell division arrest and inhibition of proliferation. 3, Semen sylvestris In clinical treatment, Semen sylvestris has been widely used to treat various tumors, and its effective anti-tumor effects have been confirmed in clinical studies, and many scholars have been conducting research on its anti-tumor mechanisms. Nina Zhang et al. established an in vivo and ex vivo angiogenesis model to study the antitumor effects of Semen nigrum. The inhibition of tumor angiogenesis and its mechanism were investigated at the cellular and molecular levels. The results of the study showed that by the action of Semen nigrum. It was shown that Semen nigrum significantly inhibited the formation of microvessels within matrix gel (Matrigel) plugs and significantly inhibited the migration of human umbilical vein endothelial cells (HUVEC); 40% of Semen nigrum-containing serum significantly inhibited the migration of vascular endothelial cells induced by cervical carcinoma HeLa cells (HeLa). The results indicated that Semen nigrum had anti-angiogenic effects both in vitro and in vivo. The study showed that Hemerocallis can effectively inhibit tumor angiogenesis, the principle of which may be related to blocking endothelial cell migration and down-regulating VEGF protein expression. 4. Chuanxiongzin Chuanxiongzin is one of the active ingredients of the Chinese medicine Chuanxiong, which has the effects of anti-platelet aggregation and improving microcirculation. In recent years, Chuanxiongzin was found to have an inhibitory effect on tumor growth and metastasis, and its effect may be related to anti-angiogenesis. Xu Xiaoyu et al. found in animal experiments that chuanxiongzin could reduce the number of foci, tumor volume and mass of lung metastases in mouse lung adenocarcinoma (Lewis lung cancer) by inhibiting the expression of VEGF in tumor cells and lowering their MVD and inhibiting their angiogenesis. It indicates that Chuanxiongzin inhibits tumor recurrence and metastasis by inhibiting tumor angiogenesis. 5.Kombu polysaccharide Kombu polysaccharide is a neutral glucan widely found in brown algae, which can be sulfated to kombu polysaccharide sulfate (, LAMS) under artificial conditions. To study the anti-tumor angiogenic effect of LAMS, Zhongping Xu and others took the chicken embryonic chorionic villi allantoic membrane (CAM) assay and found that the observed, LAMS inhibited CAM angiogenesis and was an inhibitor of bFGF attachment and bFGF-dependent cell value addition. The same study found that the inhibitory effect of LAMS on bFGF was positively correlated with the degree of LAMS sulfation. LAMS interferes with the signaling stimulation of bFGF by directly binding to bFGF so that bFGF cannot bind to its affinity site. Therefore, LAMS can be obtained to prevent tumor angiogenesis by interfering with the angiogenic signal stimulation of bFGF in tumor tissues. 6.Curcumin Curcumin traditional anti-tumor drug curcumin is a phenolic pigment component of turmeric medicinal plants such as turmeric, curcuma longa and tulip. Studies have shown that curcumin and tumor necrosis factor (TNF) can have an effective effect on the synthesis, expression and secretion of VEGF in human umbilical vein endothelium-derived cell line (EVC304) cells angiogenesis and lymphoma cell line lymphoma (Raji) cells in stromal gel. The results of the study suggested that curcumin inhibited VEGF expression and angiogenesis in Raji cells, while TNF induced angiogenesis in cellular stromal gel and promoted VEGF expression in Raji cells. Combined with Yang and Ping’s experimental study on the anti-angiogenesis of curcumin active monomer components, it was confirmed that the expression of VEGF and its receptor could be inhibited through both autocrine and paracrine pathways, suggesting that curcumin could exert its anti-tumor effect by inhibiting angiogenesis. 7, Weimannin Weimannin as a kind of tannin compound extracted from the dried rhizome of golden wheat using modern technology. Lou Jinli et al. observed that wimainin exerted its antitumor effect against transplanted lung metastasis in Lewis lung cancer hormonal mouse model through the dual action on HUVEC and metastatic lung cancer cells (PC) and reducing tumor tissue microvascular formation mechanism. Meanwhile, a related study found that wemainine inhibited the growth of transplanted tumors in mice by inhibiting tumor microangiogenesis. It indicates that wemainine manifests its effect of inhibiting tumor angiogenesis by inhibiting tumor microangiogenesis. 8.Conlet Injection Conlet Injection is a fat emulsion of Coix lacryma oil extracted from the Chinese medicine Coix lacryma. By culturing aortic rings in three-dimensional collagen gel in serum-free medium, Li Dapeng confirmed that Kanglaite injection acts on tumor cell growth process, blocking tumor cell mitosis in G2/M phase; prompting tumor cell apoptosis, inhibiting tumor neovascularization, and reversing tumor multidrug resistance. It indicates that inhibition of angiogenesis is one of the ways of anti-tumor of Conrad. 9, red cartilage and whale shark cartilage Red and shark belong to the same cartilaginous fish, relevant research results suggest that red cartilage components contain anti-tumor, inhibit angiogenesis substances. Red cartilage angiogenesis inhibitory factor-1 (DCAIF-1) was isolated from red cartilage. Xinwei Yu et al. verified its biological activity by establishing a collagenase CAM model. The results showed that DCAIF-1 could effectively prevent the migration and proliferation of vascular endothelial cells by inhibiting collagenase activity, and prevent the established tumor cells from establishing neovascular network, thus inhibiting the proliferation and migration of tumor cells. SCAIFl and SCAIF80 are two angiogenesis inhibitory factors isolated from whale shark cartilage, which were observed by Jia Fuxing et al. that they can significantly inhibit HUVEC proliferation and the inhibition occurs mainly in the s-phase of the cell cycle, suggesting that the mechanism of inhibition of endothelial cell migration and proliferation is through the inhibition of their endothelial cell DNA synthesis. 10, Gecko Gecko, also known as “Shou Gong”, “Tian Long”, is a commonly used clinical drug to fight poison with poison, and has an inhibitory effect on liver cancer and other gastrointestinal tumors, and the anti-tumor effect of fresh gecko is stronger than that of dried products. Liu Fei’s study used gecko to act on a tumor-bearing mouse model. By measuring VEGF, bFGF protein and apoptosis in tumor tissues of mice, it proved that gecko could effectively reduce the protein expression of VEGF and bFGF in tumor tissues, and at the same time could induce apoptosis of tumor cells, thus inhibiting the formation of tumor blood vessels. 11, Desmopapillin Desmopapillin: It is one of the main anti-cancer components of Zanthoxylum with poison. Research on compound Chinese medicine preparation 1. ginseng and wheat injection As a common drug in clinical practice, ginseng and wheat injection has the therapeutic effect of benefiting qi and nourishing yin. The immunohistochemical assay showed that ginseng injection could effectively reduce the expression of proliferation nuclear antigen (PCNA) and MVD in tumor tissues, and Woxingde et al. confirmed that ginseng injection could inhibit the content of matrix metalloproteinase-2 (MMP-2) and the expression of bFGF in tumor tissues of S180 solid tumor in mice. It indicates that ginseng mai controls tumor recurrence and metastasis by inhibiting tumor angiogenesis. 2.Tao Hong Si Tang Xu Xiaoyu et al. found that this formula could effectively inhibit CAM angiogenesis, the growth of B16 melanoma and the proliferation of ECV304 cells, and reduce MVD in tumor tissues. its anti-tumor mechanism of action may be related to the inhibition of endothelial cell vascular endothelial growth factor 2 (KDR/FLK-l) expression, and its anti-tumor effect inhibits anti-tumor angiogenesis The anti-tumor effect is probably related to the inhibition of endothelial cell vascular endothelial growth factor 2 (KDR/FLK-l) expression and its anti-tumor effect on anti-tumor angiogenesis. 3.Turtle shell decoction pill Turtle shell decoction pill is a famous formula for softening and dispersing hard nodules, which is derived from the book “Jin Kui Yao” written by Zhang Zhong Jing, a doctor in the Eastern Han Dynasty. Chen Dali et al. established an animal model of tumor-bearing mice and used turtle shell decoction pill for in vivo intervention. By observing the expression of VEGF in tumor masses of tumor-bearing mice, it was found that it had a significant inhibitory effect on the expression of VEGF in tumor cells in mice. 4.Lung Yan Ning Lung Yan Ning is composed of Astragalus membranaceus, Atractylodes macrocephala and Toadstool, etc., to benefit Qi and nourish essence, detoxify and disperse cubes. Through the study of clinical efficacy, Xu Zhenye et al. found that lung Yan Ning has the effect on the secretion of VEGF and the effect of anti-invasive metastasis of advanced non-small cell lung cancer. Reducing VEGF secretion and inhibiting neovascularization may be one of the mechanisms of anti-lung cancer growth and metastasis. 5.Aikang granule Aikang granule is made of Curcuma longa, Cichorium sanctum and ginseng, among which Cichorium sanctum contains “colchicine”, an anticancer ingredient that can inhibit cell mitosis and stop the proliferation of tumor cells. Dong Hailing et al. found that Cichlidium may have the effect of inhibiting tumor angiogenesis. The alcoholic extract of C. sanguinis can strongly inhibit bFGF-induced HUVEC proliferation in vitro, and also inhibit CAM angiogenesis in vivo. Elemene is the main component in Curcuma longa, and by studying the effect of elemene on the growth of hepatocellular carcinoma cell line SMMC-772, Xu Xuejun et al. found that elemene induced apoptosis in hepatocellular carcinoma cells probably due to increased expression of human oncogene (p53), and decreased expression of oncogene B lymphocytoma-2 gene (bcl-2) and proto-oncogene (c-myc). Feng Gang et al [32] used curcuma oil injection for treatment to establish a sarcoma of S180 in a tumor-bearing mouse model, and the results confirmed that curcuma oil injection could significantly inhibit the expression of MVD and VEGF and bFGF in the tumor. It was suggested that curcuma could promote the proliferation, adhesion, migration and lumen structure formation of endothelial cells and up-regulate the expression of certain angiogenic factors through down-regulating bFGF protein channels. Related studies have shown that tumorigenesis, metastasis, recurrence and prognosis are closely related to tumor angiogenesis. The anti-tumor angiogenic drugs of Chinese medicine have gained increasing attention because they are less likely to develop drug resistance, do not have toxic side effects such as gastrointestinal reactions or bone marrow suppression, which are common with traditional chemotherapy drugs, and have broad-spectrum anti-tumor effects. Anti-tumor angiogenic drugs have good application prospects. Chinese medicine has its special advantages in the treatment of tumors, whether it is to cooperate with radiotherapy, enhance the efficacy and reduce toxicity, or to relieve clinical symptoms, improve the quality of survival, prevent tumor recurrence and metastasis, and prolong survival time. Because of the various mechanisms and pathways of tumor angiogenesis, it is difficult for a single drug to completely block tumor angiogenesis. The multi-taste, multi-generalization and molecular diversity of traditional Chinese medicine characterize the multi-target action of traditional Chinese medicine, and the combination of Chinese medicine may completely block tumor growth by inhibiting tumor angiogenesis, which will become one of the hot spots of anti-tumor research of Chinese medicine.
