Aleitinib, whose full name is Aleitinib Hydrochloride Capsules, is an antineoplastic agent that usually does not require a dose adjustment for mild to moderate hepatic impairment and usually requires a dose reduction for severe. Aleitinib is a highly selective and potent ALK and RET tyrosine` acid kinase inhibitor. In non-clinical studies, inhibition of ALK tyrosine kinase activity blocked activation of the downstream signaling pathways STAT3 and PI3K/AKT, inducing tumor cell death (apoptosis). It can inhibit tumor growth. Aleitinib is indicated for the treatment of patients with locally advanced or metastatic non-small cell lung cancer that is positive for interstitial lymphoma kinase (ALK). According to the insert, no dose adjustment is necessary for patients with mild (Child-Pugh class A) or moderate (Child-Pugh class B) hepatic impairment. For patients with severe (Child-Pugh class C) hepatic impairment, the dose should be 450 mg orally twice daily (900 mg total daily dose); the drug may be discontinued if the impairment is severe.
