Usually, sperm viability can be tested by doing eosin staining and hypotonic swelling test. 1. Eosin Staining: It is to prepare eosin into a dye with saline, mix a drop of fresh semen with a drop of eosin dye on a slide, cover it with a coverslip, and examine it under a light microscope after 30 seconds. Usually, normal spermatozoa are not stained, only dead ones are. If a large number of sperm stained red are found under the microscope, it indicates that the sperm viability is relatively low; if the sperm survival rate is ≥58% it is normal, otherwise it may affect normal fertility. 2. The hypotonic swelling test removes the more viable sperm from the inactive sperm, and ultimately detects whether the sperm are abnormal. A sperm hypotension test of ≥60% is considered normal.
