How should melanoma be diagnosed?

  There are some tests that must be done when you have melanoma, because these melanoma tests are important for the treatment of melanoma in order to understand the condition of melanoma patients more clearly.  How is melanoma detected?  1. When the antiserum dilution was 1:400, 82.14% showed positive reaction when measured by Vacca double PAP immunoenzyme labeling method. This is a common test method for melanoma.  2. Pigmentogen test, oxidation of melanogen after renal excretion, which makes the urine dark brown, called black urine, is of great significance for the examination of melanoma. If iron chloride, potassium dichromate and sulfuric acid are added to the urine, it can promote its oxidation, then add sodium nitrate, the urine will be purple; if acetic acid is added first, then sodium hydroxide, the urine will be blue.  3.Anti-human melanoma serum does indirect immunofluorescence labeling of melanoma tissue, and the highest positive rate can reach 89% when the antiserum is diluted to 1:2, which also belongs to the common examination method of melanoma.  The examination methods for malignant melanoma include: 1. Pathological examination is a reliable means to examine melanoma and is also the main method for differential diagnosis. When the clinical conditions allow, the entire thickness of the tumor and the infiltrated tissues in all layers of the skin should be removed and biopsy should be performed to obtain pathological diagnosis. Needle aspiration, scraping, clamping, cutting or chiseling should be avoided to prevent the spread of lesions.  2.Electron microscopy The melanosomes of malignant melanoma at different stages of development can be observed, which are characteristic and can be identified with other tumor cells and phagocytic melanocytes. The observation of immunoenzymatic labeling (S-100), monoclonal anti-HMB-45 and flow cytometric DNA analysis are of diagnostic value in confirming the diagnosis of malignant melanoma.  3.X-ray examination Chest X-ray should be performed for possible, asymptomatic lung metastases. If it suggests metastasis, lung tomography or CT scan should be applied to confirm melanoma.
