The method of urine culture includes the collection and delivery of urine specimens, specimen acceptance, and the method of culture. Finally, we observe the results as well as write the report. 1, collection method: drink less water before going to bed, preferably morning urine, and wash the perineum after waking up to ensure the cleanliness of urine. Males should wash the foreskin as well as around the urethra thoroughly. Women should turn the labia majora over and clean it. Then access 10-20 ml of urine into a sterile container. 2, culture method, (1) ordinary culture: the container for collecting specimens will be gently rotated and mixed, with a quantitative inoculation ring to take 1μl of urine to be coated with inoculated blood plate and Chinese blue plate respectively, culture temperature at 35-37 degrees Celsius, time in a day and a half to two days, and then observe the results of the urine test. For patients taking antibiotics or specimens from suprapubic cystocentesis an additional 10μl inoculum is required. (2) Special culture: For suspected anaerobic infection, a chocolate plate should be added and incubated in a 5% CO2 environment for 48 hours. (3) Factors affecting the culture: The amount of bacteria increases when urine is left for more than 1 hour. The necessary cleaning work before collection is not done, and the middle section of urine is not clean. The method of urine culture is so essential skills for the examiner and needs to be fully mastered.
