The time required for specimen fixation depends on the size of the specimen, generally 4-6 hours for small specimens, and 18-24 hours or longer for large specimens; 3, specimen delivery and specimen numbering and registration, specimen observation and sampling, the specimen delivered for examination, regardless of size, should be carefully observed and recorded for its morphology, size, color, surface and section changes, the presence of abnormal lesions and tumors, and the relationship with the surrounding. The size of the tissue taken for sectioning is 1.5×1.5cm, and the thickness is between 0.2-0.3cm; 4. Preparation of tissue sections: nowadays, it is done in automatic dehydration machine and staining machine. The procedure includes fixation, dehydration, transparency, wax dipping, embedding, slicing, staining, sealing and labeling; 5, after the pathology preparation: the tissue sections are handed over to the pathologist, and the technician checks the number of sections with the doctor, makes a good handover and records; 6, reading: the sections are observed under the optical microscope, and the diagnosis of various diseases is made through the analysis and identification of the lesion tissue and cell morphology, and then combined with the naked eye and clinical information. However, for some difficult and rare cases, it is also necessary to make auxiliary diagnosis through histochemistry, immunohistochemistry, electron microscopy or molecular biology technology on the basis of the above-mentioned routine examination; 7, pathological diagnostic report issuance: the issuance of pathological diagnostic report, to make a good handover with the clinician, in the absence of special examination, from the receipt of specimens to the issuance of the pathological report, the process of biopsy takes about 5-7 days.
