How to properly retain urine specimens

What is the correct way to keep urine specimens? The urine specimen should be preserved as long as you follow the following methods and precautions, your urine specimen will meet the requirements of urine specimens, and avoid repeated testing due to urine specimens failed, delaying the diagnosis and treatment of the situation. 1, the specimen is best to clean the urethral opening and vulva before retention, the retention process should avoid contamination by menstrual blood, leukorrhea, semen, feces, etc. mixed in. 2.Fresh and clean middle urine should be retained. A urination process can be divided into anterior, middle and posterior segments according to the sequence of urine outflow, because the anterior and posterior segments are easily contaminated, so the middle segment of urine should be retained. Count three seconds from the beginning of urination (note that you should urinate continuously without interruption), then immediately pick up the urine, at this time intercept the middle segment of the urine, retain about 10 to 12 ml as a specimen for testing, to such as disposable urine tube inside. 3, the container should avoid the presence of drugs and a variety of disinfectants to prevent the destruction of a variety of organic fraction form and hormonal activity. According to the requirements of different items, choose whether to use sterile containers, and whether they need to be kept refrigerated. 4, 24-hour urine should be strictly controlled for all urine for 24 hours. Because the concentration of various substances in human urine varies at different times, for example: electrolytes, catecholamines, 17-hydroxysteroids, etc., have the lowest content in urine in the morning and higher content in the afternoon, so all urine within 24 hours must be strictly retained. Note that the urine specimen should be placed in a cool and ventilated place during urine collection to reduce microbial growth and maintain a constant urine PH. 5. The best time for specimen delivery is within 30 minutes, and the longest time should not exceed 2 hours, otherwise it will cause: (1) bacterial growth and reproduction in the urine; (1) urea enzymolysis, so that the urine PH value increases and the organic fraction is destroyed (tubular decomposition, red blood cell lysis); (2) glucose is degraded, so that pathological glucosuria disappears; ③bacterial bacteriophage protein and pathological proteinuria interfere with each other. (2) The chemical components in urine (urobilinogen) can be denatured and decomposed by prolonged storage of urine; salts in urine interfere with microscopic examination by precipitating crystals due to prolonged storage, thus affecting the judgment of urine results. Ensuring the quality of the specimen before the test is the first step in improving the accuracy and reliability of the test results, which requires your full cooperation.
