Inhibition of experimental atherosclerosis in rabbits by the worm-like blood-activating drug Shuanglongwan

  Shuanglong pill is derived from the classical formula Stop Spasm, which consists of Di Long, Quan Scorpion and Centipede, and has the effects of pacifying the liver and submerging the Yang, searching the wind and picking up the ligaments, activating the blood and fixing the pain, and preliminary studies have confirmed the effect of regulating the PGI2/TXA2 balance. myocardial ischemic effects are related to endothelial protective effects []. The aim of this study was to investigate the effects of shuanglongwan on smooth muscle cell proliferation and migration at the cellular and subcellular levels by establishing a rabbit atherosclerosis model with light and electron microscopy, and to provide a theoretical basis for its further clinical application.  1. Materials and methods 1.1 Experimental drugs and instruments Cholesterol: Sigma (USA) Shanghai distribution; lard: purchased from the market. Kaipotong: produced by Sino-US joint venture Shanghai Schweppes, lot number: 0108031, 12.5mg/capsule. Shuanglong pill: purchased from the pharmacy department of Shanghai Shuguang Hospital, 0.5g per capsule; pathology microtome (LEICA RM 2165, made in Germany); spreading and baking machine (CS-IV, made by Xiaogan Electronic Instrument Factory); optical microscope photography system (OLYMPNS, BX60, made in Japan); H-600-4 transmission electron microscope (made by Hitachi, Japan); electron microscope uptake system (PANASONIC; model number: 0108031). system (PANASONIC; model MV-CP410, made in Japan).  1.2 Animal models Male New Zealand Rabbits (provided by the Experimental Animal Center of Shanghai Shuguang Hospital) weighing 2.0-2.5 kg were randomly divided into four groups of six rabbits each and housed in single-group cages. Group III: Captopril control group, 1g of albumin was injected early in the morning of the modeling day, then fed a diet containing 1g of cholesterol and 3% lard per day for 12 weeks with unlimited water. Group IV: Shuanglong pill group: 1g of albumin was injected in the early morning of the modeling day, then fed a diet containing 1g of cholesterol and 3% of lard every day, and each animal was given Shuanglong pill (2.52g/kg/d, 10 times the adult dose) for 12 weeks by gavage, and water was not limited.  1.3 Observation items 1.3.1 Light microscopic examination The free aortic segment was executed at the end of the experiment, and the free aortic segment was cut open along the long axis to observe visually whether the inner wall of the artery was smooth and whether there was AS plaque. 1cm arterial segment at the aortic arch was taken and fixed in 10% formalin buffer, HE stained and paraffin embedded for light microscopic examination.  1.3.2 Electron microscopic examination After air embolism, the aortic trunk was quickly removed and immediately put into 2.5% glutaraldehyde for pre-fixation at 4℃ for 2h, fixed with 1% osmium acid for 2h, placed in 70% alcohol in the refrigerator overnight, and embedded with epoxy resin 618 to prepare ultra-thin sections. Electron microscopy was observed.  1.4 Statistical treatment SPSS10.0 statistical software, independent t-test, was used and expressed as mean ± standard deviation.
