For patients with metastatic kidney cancer after treatment failure with sorafenib or sunitinib, everolimus was approved by the US FDA in 2009. Everolimus is an orally administered mTOR inhibitor. The relevant evidence was derived from data from a phase III clinical trial for the treatment of metastatic kidney cancer that had failed previous targeted therapy, with a median progression-free progression interval of 4.9 months for everolimus compared to placebo or blanket therapy. Axitinib is an emerging multi-targeted receptor complex kinase inhibitor that has been approved by the FDA for second-line treatment of advanced kidney cancer in early 2012. A phase III clinical trial comparing axitinib with sorafenib for metastatic kidney cancer that has failed previous targeted drug therapy found that axitinib was superior to sorafenib. Of these drugs, axitinib and pazotinib have not yet been available in China, but everolimus was launched in China in 2014 for patients who had failed prior treatment with sorafenib or sunitinib. Aggressive treatment of metastatic lesions may also help relieve painful symptoms of kidney cancer patients and may prolong survival.
