With the promotion of the 2006-2011 NCCN Guidelines for the diagnosis and treatment of central nervous system tumors (NCCN Guidelines) and the Chinese Consensus on the diagnosis and treatment of malignant glioma of the central nervous system (Chinese Consensus), the number of patients who have relapsed after completing the standard treatment regimen of temozolomide has gradually increased, and there is no uniform opinion on how to treat these patients. In addition to the 1week on/1 week off and 21/28 regimens of temozolomide, the 28/28 regimen has attracted attention. In 2010, JCO published high evidence-based results from a multicenter clinical trial of 11 Canadian institutions participating in a continuous drug-density regimen of temozolomide for the treatment of recurrent high-grade glioma. The 6m-PFS of rGBM patients in the rechallenge group was 35.7%, and the 1y OS of patients enrolled in the treatment was 28.6%; and the toxicities of the drug during treatment were within acceptable limits.
